Objective: To study the effect of angiotensin II on m-RNA expression of transforming growth factor P 1 (TGF-β1) -and secretion of extracellular matrix(ECM) of cultured rat glomerular mesangial cells, and the suppressive effect of Chinese Herbal xiao~ban~yu-shen-ji (XBYS) on them, to investigate the cellular and molecular mechanism of XBYS in preventing mesangial sclerosis.Methods:1. XBYS pharmacological serum, cordicosteroid pharmacological serum and shen yan shu pharmacological serum were got by serum pharmacology 2. the mesangial cells were cultured by microcell culture method 3. m-RNA expression of TGF- β1 were measured by RT-PCR 4. The secretion of fibronection (FN).and collagenlV examed by enzyme-linked immunosorbent assay( ELISA)Results: 1. angiotensin II increased m-RNA expression of TGF- β1, biosynthesis and secretion of FN and collagenlV;2. XBYS inhibited the expression of TGF-β1 m-RNA, XBYS also inhibited the secretion of FN and collagenlVConclusion:XBYS can significantly inhibit the secretion of FN and collagenlV , m-RNA expression of TGF- P 1 in vitro, which may be the mechanism of XBYS in prevention and treatment of glomerusclerosis in mesangial sclerosis.
