HCC is one kind of common and highly malignant tumor existing in east south Asia. There are at least 110,000 persons died of HCC in China every year, occupying 40% of total in the world.The pathological data indicates that many elements have been involved in the generation and progression of HCC. Our group mainly focused on the gene expression pattern of HCC, and we have found some HCC "associated" genes by the analysis of "RDA" and "cDNA array"data, most of which are known genes, studying of them has implied some information for the reasoning of HCC. Some of them are novel gene , some are even nothing to know, which add difficulty for us to analysis. Being novel genes, it is necessary and worthy to study the function, so we selected one and named it SVH as the protagonist of my work. SVH belongs to a large protein family consisted of Alex-1, 2, 3, BM-017and some other unknown proteins, which has high homogeneity with SVH. Till now, their function has not yet been reported, so to identify the function of SVH is significant to reveal the function of such protein family. Basing on the data bank of NCBI, we cloned this gene by RT-PCR. SVH has a completed ORF of 1119bp.Nortern Blot data indicate that SVH has three different transcripts, the shortest one is about 2kb.SVH expressed broadly in kidney, intestine, stomach, cortex and spinal cord etc in human fetus of 20 weeks old. We are the first to find that SVH has five isoforms, and cloned four of them, another one appears in EST databank. These isoforms have tissue distribution difference; it is interesting to find that four of them can exist in one cell. We roughly locate SVH in the Endoplasmic reticulum bythe experiment of immunostaining using the antibody prepared by ourselves, which is also confirmed by the data from SVH-GFP fusion protein locating analysis of each isoform.
