As one of emerging microorganisms, infection caused by Escherichia coli O157:H7 has become a worldwide public health problem, including China. Epidemiological studies suggest the treatment of hemorrhagic colits(HC) by O157:H7 with antibiotics may lead to the enhancement of shiga-toxin (SLT) producing , further increase the possibility of He patients developing hemolytic uremic syndrome .It is necessary that searching a remedy ,which can therapy the illness induced by Escherichia coli O157:H7 effectively ,avoiding side effect at the same time .In this research, we adopted isolated lactobacillus acidophilus as the drug for therapy . A streptomycin-treated BALB/c mouse was used.The srain Escherichia coli O157:H7 is 88-2364, standard strain in China. It was fed orally by calibrated droppers. The test is divided into 5 groups: preventive group, lactobacillus acidophilus fed 6 hours after feeding 88-2364, lactobacillus acidophilus fed 12 hours after feeding 88-2364,positive control group, negative control group.Observing clinical symptoms of the mouse, including neurological system symptoms, such as lethargic and anorexia, hallux paralysis ,note the death rate .Examine live O157:H7 of stool by a rapid immunochromatographic test everyday. Seven days postfeeding or when signs of death appears, animal was sacrificed and necropsy. Tissues specimens was collected for histological examination. Exact the toxin of stools, culture vero cell, examine the level of toxin by Cytotoxicity detection kit (LDH).The result is that lactobacillus acidophilus can decrease the mortality rate, live O157:H7 of stools. The level of Shiga-toxin in the stools descend distinctly. Histological examination suggest that infiltration of inflammatory cell descendobviously. Debris and lysis of nucleolus of renal convoluted tubules alleviate.These results show that lactobacillus acidophilus may have the potential to be used for the treatment of patients with HC caused by Escherichia coli O157:H7.
