Adaptin 2 is a key part of clathrin-coated vesicles (CCVS), which participates in protein transduction from membrane to the cytoplasm. Hybridizing human adult testis cDNA microarrays with human adult and 6 months embryonic testis cDNA probes, we identified a novel splicing isoform of adaptin protein 2β (Ap2β), named Ap2β-NY, of which mRNA expression level was 3.62-fold stronger in human adult testes than fetal testes. After sequencing, it's whole sequence, containing 3457bp, was obtained and deposited in the GenBank(accession number, AY341427). Ap2β-NY encode a protein of 880 amino acids with presumed molecular weight of 98.12KDa kDa and isoelectric poin of 5.01 and was mapped to ql 1.2-q 12 regions of chromosome 17. Predicted protein motif analysis of Ap2β-NY revealed that it had all the three motifs of Ap2β and is 57 amino acids shorter in the N-terminal region than Ap2β The deleted region is hydrophilic and likely exposed on the surface of the protein, which, in general, was the position of protein interacting with others. By use of RT-PCR, Ap2p-NY was indicated to highly expressed in adult testis and spermatozoa while weakly expressed in embryonic testis, which validated the result of hybridization at the same time.
