Objective: To investigate the analgesic effect of Toutongping(a Chinese herbal drug granules); To investigate the pathogenesis role of calcitonin gene-related peptide(CGRP) in migraine; To get the control mechanism of Toutongping to the level of CGRP in the blood of migraine rats.Methods: The mice are divided randomly into five groups: Normal control group, Toutongping high, middle, lower groups; Zhengtian- wan group. Give the drugs for three days, then measure the level of algesthesia reflection in every group after they are stimulated by hot, electron and glacial acetic-acid; The rats are divided randomly into five groups : Normal control group; Model control group; Toutongping high, middle, lower groups; Zhengtian wan group. The model of migraine rats are produced by hypodermic injection of nitroglycerin. Give the drugs for six days and on the seventh day produce the model of migraine rats. After four hours, the content of CGRP is detected by radio-immunity analysis method.Results: Compared with normal group, the drug groups can decrease the level of algesthesia reflection of mice; The CGRP content of the blood in the orbital veins: The content is constantly kept in lower level in the normal rats. The content of model groups is significantly increased (p<0.01); The contents of drug groups are approached to that of the normal group, but compared with the model group, they are significantly reduced (p<0.01).Conclusions: Toutongping has the effectiveness of analgesia; The excessive expression of CGRP in the experimental rat vein blood with migraine may be one of molecular mechanisms to trigger migraine; the controlling and regulating of toutongping to express CGRP may be one of the mechanisms that effectively prevent and treat migraine.
