Juncus effusus L.(Fam. Juncaceae) sweet, tasteless, slightly cold. Distribute to the heart, lung and small intestine meridian, has the action of eliminating excess fire in the heart, inducing diuresis, was used in treating restlessness and insomnia etc. These actions are the same as the action of central inhibition such as sedation and hypnosis. This study aimed to verify its sedative and hypnotic effect and screen out active components through general study method of chemistry and pharmacodynamics.This dissertation includes two parts.Part 1, Overview: Overviewed commonly used sedativehypnotics and their pharmacological research methods ; the research progresses about chemical composition and pharmacological action of Juncus effusus L, 38 references were cited.Part 2, Experiments: First, 95% ethanol, the best extraction solvent was determined through the results of pharmacodynamic experiments about the different extracts. Then, the whole herb of Juncus effusus was extracted with 95% ethanol. The ethanolic extract was further separated with petroether, chloroform, ethyl acetate and methanol in turn to give four fractions. The ethyl acetate fraction was proved to be the most active fraction, which was further seperated into several fractions with column chromatography. Then, more centralized active fractions 102 and 104 were screened out by pharmacodynamic experiments.Systematically studied chemical compositions of 102(chemical compositions of 202 were separated in earlier period), isolated many components. Based on the spectra data(1D,2D NMR, UV, MS etc.), 20 of them were identified, involved 6 species, included 6 flavonoids: luteolin(A), 7,4'-dihydroxy-5,3'-dimethoxyflav- one(C), 7,3',4'-trihydroxy-5-methoxyflavone(E), 5,7,4'--trihydroxy-3'-methoxy- flavone(L), tricin(P), quercetin(S); 5 dihydrophenanthrenes or phenanthrenes which contain biphenyl construction: 7-carboxy-2-hydroxy-1-methyl-5-vinyl- 9,10-dihydro-phenanthrene(J), 7-carboxy-2-hydroxy-1-methyl-5-vinyl-phenanth- rene(N), 8-carboxy-2-hydroxy-1-methyl-5-vinyl-9,10-dihydrophenanthrene(Q), effusol(T), dehydroeffusol(U); 5 organic acids: protocatechuic acid(D), caffeic acid(F), p-coumaril acid(K), p-hydroxybenzoic acid(M), ferulic acid(R); 2 sterols:β-sitosterol(I), daucosterol(B); 1 anthraquinone: emodin(G); 1 fatty acid ester: hexadecanoic acid ethyl ester(H). Among them, components C, E, L, P, N, D, F, M, R, G, H were firstly isolated from this plant, component N is a new component.Finally, the sedative activity of major isolated biphenyl construction contained components J, Q and U; other biphenyl construction contained components bicyclol and...
