Introduction: Dan-shen root is one of traditional Chinese herbs which can promote blood circulation and to remove blood stasis. With the development of medicine, many studies also found that Dan-shen root plays an important role in the management of cardiovascular disease and cerebrovascular disorders. Dan-shen root has different kinds of chemical compositions, which have different functions respectively. The pharmacological study on the Magnesium Lithospermate B (LSA-B) has been less reported because of the complex technique of obtaining the water-dissovling extracts. The main aim of this work is to investigate the machnism of LSA-B on cardiovascular system and to provide some experimental evidence for further study.Objective: To study the effect of LSA-B on the proliferation of human aortic endothelial cells (HAECs) and the possible protectively effect on the HAEC injured by the free radical.Methods: 1) Cell culture: HAECs were purchased from Cascade Company (USA) and were cultured with the medium 200. The cells of Passage 3 to Passage 6 were used in the experiment. 2) MTT test for the evaluation of cell proliferation: the effects of different concentration of Magnesium Lithospermate B on HAEC proliferation were observed. 3) The free radical injury to HAEC: Glucose Oxidase model was used in the experiment.Results: Cell damage was observed in the high concentrations of 1mg/mL 0.5mg/mI. There is not injurious effect in the concentrations below 0.2mg/ml on the cell proliferation. An evident protective effect of Magnesium Lithospermate B on HAEC injured by the free radical was observed.Conclusions: There is a large gentle concentration range for using Magnesium Lithospermate B in the management of vascular disease. They have no toxic effect on EC proliferation, and show a protective effect on HAEC against the free radical injury.
