Cyclooxygenase Inhibitors And Ovarian Tumors

Object:(1)To investigate the expression of COX-1 and COX-2 in ovcarian carcinoma.To evaluate the correlation between the COXs and clinico-pathologic variables in these ovcarian carcinoma patients.(2)To study the expression of COX-2 of celecoxib,a selective cyclooxygenase-2 inhibitor,and aspirin,a non-selective cyclooxygenase inhibitor on two ovcarian carcinoma cell lines in vitro.(3) To study the inhibitory effects of celecoxib and aspirin on two ovcarian carcinoma cell lines in vitro.Methods: (1) The expression of COX-1,COX-2,were examined by immunohistochemistry method and tissue chip method in 10 cases of normal ovcarian tissues,10 cases of benign ovcarian tumors,8 cases of ovcarian borderline tumors and 39 cases of ovcarian carcinomas.(2) The expression of COX-2 were analyzed by immunocytochemistry. (3)The effect of cell cycle and cell apoptotic rates were analyzed by flow cytometry(FCM).Results: (1) The positive expression rates of COX-1 have no significant difference between ovcarian carcinoma, borderline tumors, benign tumors and normal ovary(P>0.05).The COX-2 protein expression in ovcarian carcinomas and borderline tumors were statistically higher than those in benign tumors and normal ovary(P < 0.05),but there was no significant difference between ovcarian carcinomas and borderline tumors(P>0.05).The expression of COX-1 in ovcarian carcinoma has no correlation with clinical stages,grades and metastasis of lymphatic nodes; The expression of COX-2 in ovcarian carcinoma has no correlation with clinical stages and metastasis of lymphatic nodes; but it has a correlation with grades(P<0.05).(2) By immunocytochemistry,When incubated with aspirin(≥5mmol/L)/celecoxib(≥25μmol/L),it was found that the positive expression rates of COX-2 were statistically declined of two ovcarian carcinoma cell lines in vitro(P<0.05).(3) By FACS,it was found that the cell cycles of 3AO and OVCAR3 cells were blocked by either aspirin and celecoxib,with the increasing of concentration: the G0/G1 phase ratio of 3AO/OVCAR3 were increased,the S phase ratio were inhibited and the G2～M phase ratio were not effected significantly.When incubated with aspirin(≥5mmol/L)/celecoxib(≥...
