Hypoxic pulmonary hypertension(HPH), characterized by elevated pulmonary blood pressure, pulmonary vascular remodeling, and right ventricular hypertrophy, is a common complication of cardiopulmonary disease; Without intervention, HPH has a progressive downhill course, leading to right ventricular failure and death. As the pathogenesis remains poorly understood,there is still no ideal treatment for HPH and alternatives are still required.It is very important to elucidate the mechanism of HPH and search for new classed of treatment for this debilitating disease. It is evident that the process of HPH is a complicated one with several cellular and molecular mediators interacting in concert, however, the mechanisms of initiation and maintenance of HPH remain obscure.Great importance has been attached to the contribution of Rho/Rho kinase signaling to cardiopulmonary disease.Evidence accumulated that this signaling had protective effects on cardiopulmonary injury,however,the mechanisms underlying are to be elucidated.In this study, the development of vascular changes in hypoxic pulmonary hypertension rats was likely initiated by endothelial cell injury was reported.In parallel studies,the expression of Rho kinase gene and protein were examined by semi-quantitative reverse transciption and polymerase chain reaction(RT-PCR) and western blot within intra-pulmonary vessels,and the effects of Rho kinase inhibitor fasudil on HPH rats were investigated so as to evaluate the therapeutic value and elucidate the mechanism of pulmonary hypertension.Part I: The effects of Rho kinase inhibitor fasudil on pulmonary hypertension and vascular remodeling in hypoxic ratsObjective: To establish the rat model of hypoxic pulmonary hypertension (HPH),discuss the pathological mechanism of HPH and the effects of...
