Part I Protective effects of Edaravone on hypoxic-ischemic brain damage in newborn rats.Objective: To investigate the protective effects of Edaravone on hypoxic-ischemic brain damage in newborn rats.Methods: 117 7-day-old newborn SD rats were randomly divided into hypoxic-ischemic model, high dose Edaravone -treated , low dose Edaravone -treated and sham groups. HIBD was established in the hypoxic-ischemic model and Edaravone -treated groups. Edaravone or NS was injected intraperitoneally 0 min after HI insult, and they were used for consecutive 5 days. The contents of MDA and SOD in the brain were assessed at 24h, 48h, 72h respectively after HI insult. The hippocampal changes were observed by electron—microscope. The influence of Edaravone on apoptosis of neurons in the cortex and hippocampus at 72h after HIBD was evaluated by means of TUNEL. The expression of Caspase-3 mRNA in the cytosol of the cortical neurons was assessed by in situ hybridization(ISH) at 72h after HI insult.
