BH is a vasoactive drug with a variety of pharmacodynamic properties. Importantly, it seems to improve nutritional blood flow in ischaemic tissue of patients with peripheral and/or cerebral vascular disease by a combination of pharmacological effects, and it was widely used in the treatment of peripheral vascular disorders.The thesis was aiming at preparing of SRT to improve the patient compliance, maintain relatively constant plasma concentration, by using hydroxypropylmethylcellulose (HPMC) as basic matrix material with a combination of two technologies—coating technique and twice wet granulation process.According to the literature, UV spectrophotometry was developed for in vitro assay of physicochemical properties, content and drug release. High-performance liquid chromatography (HPLC) was applied in the determination of plasma concentration in Beagle dogs.In the preformulation researches the physicochemical properties of BH were investigated. Different crystal forming in various conditions was identified by melting point and DSC, neither polymorphs nor solvate was discovered. In addition, the chemical stability of solid BH was observed, the result of which indicated that temperature, light, moisture and oxygen had little effect on it.Basing on the studies of the influence of formulation and manufacture on the release of SRT, optimal formulation modified to release drug over 16hr was obtained by orthogonal design test.
