| Objective:The pulmonary interstitial fibrosis rat model was formed by Bleomycin injection via trachea in order to investigate the mechanism and pathway of pulmonary interstitial fibrosis ameliorated by Chinese crude drug prescription,"pneumonia NO.1"(PN) and to observe its therapeutic effect of treating pulmonary interstitial fibrosis, expecting that this research can make a breakthrough in treating this disease with TCM, and providing sufficient theoretic and experimental proof for clinical treatment. Methods:Sixty-four male Wistar rats were divided into four groups randomly: the control group, the model group, the PN group and the hormone group. Animals of the control group were injected with isotonic Na chloride through trachea; those of the other groups were injected with Bleomycin in the same way to establish the pulmonary interstitial fibrosis rat model. After modeling, the PN group were gavaged with PN in the dose of 1ml/100g, the hormone group were subcutaneouly injected with cetacort in the dose of 2mg/100g, the model and control group were gavaged with isotonic Na chloride one time per day, all groups were treated for 4weeks. During the process, the general condition such as food and drink intake, body weight, vitality, movement, fur color, respiration, etc. were observed. The animals were executed in batch 7 and 28 days later. The rats' left lungs were fixed with formalin, and the slices were disposed in routine method, dyed with Hematoxylin-Eosin, bitter acid-rubrum scarlatinum, and observed with computer image analysator. At the 7th day, the rats' right lungs were treated by bronchoalveolar lavage with sterilized isotonic Na chloride (37 ℃ ), and the bronchoalveolar lavage fluid were retrieved to obtain alveolar macrophage. At the 28th day, primarily culture the macrophages from the right lungs interstitial substance. The cultured cells were stained with nucleic acid hybridization in situ to observe the TGF- β1 mRNA expression. Results:1. Rats of the normal control group were active, agile, muscular, aggressive, with bright fur, good appetite, growing body weight, normal respiration, urine and stool, and no rat died until execution in this group; As for the model group, the animals were dispirited, lazy, slow, crouching, with dry, dark, or inverse fur; the food intake and defecation reduced. At the 28th day, rats of this group were notably thin, idle compared with the other groups and coughed with asthma.
