Atherosclerosis is a kind of complex multifactorial disease. Many complicated factors interaction and interrelated biological processes contribute to atherogenesis. Study of its mechanisms can provide available preventive measures and clinical therapy strategies. At present study, apoE-/-/LDLR-/-/Leprdb/db treble-gene mutant mice were established in our lab, and used for study the mechanisms of polygenetic disfunction in dyslipidemia as well as atherogenesis. The relationship between treble-genetic mutation and hyperlipidemia and hyperlipoprotein, following atherosclerotic pathological changes in the mutant livers were investigated. The influence of high fat/cholesterol diet induction on the development of hyperlipidemia and atherosclerosis was studied.In this paper, another factor with high sugar drinks (prepared by 20% sucrose solution) is added to the investigation of how diet effect the concentration of TC, TG, HDL-C, LDL-C in blood serum of treble fatty metabolism genes mutant mice and the pathology changes in the livers of treble-gene mutants. The preparation and detection of allicin also discussed in this experiment. Based on these findings, the influence of allicin solution in induction on the development of hyperlipidemia and atherosclerosis was then studied compared with placebo of distilled water.The following result can be got from this experiment:1. The best extraction of allicin can be got when using 90% alcohol solution as extracting agent. And using HPLC to detect the concentration of allicin is best and suitable way in research and manufacture.2. ALO mice received allicin by intragastric administration. The level of TC and LDL-C are decreased compared to those mice received distilled water. But the level of TG and HDL-C are not decrease significant when given allicin by intragatstric administration.3. ALO mice received allicin by intragastric administration. The body weight have no visible Changes compared to those mice received distilled water. And in another way, the appetite of ALO mice received allicin is not change obviously except the first week.4. ALO mice received allicin by intragastric administration. When fed with normal food, the liver apomorphosis is obviously changed compared to those mice received distilled water. But when fed with high sucrose /fat/cholesterol diet, the efect of allicin is go to show up, and the fat drop in liver cell is became more small and less than those mice given by distilled water as placebo.
