| The incidence of cholelithiasis in adult is 3.5% to 9.0% in China. Surgery is a primary treatment with choledocholith and hepatolithiasis, which consiste of pigment gallstones. However, it is very difficult in operation, and the rate of lithobiotis and lithogeresis is still high after operation. So, it is significant to discover an effective oral drug to treat choledocholith and hepatolithiasis. As a traditional Chinese medicine compound, Baogan Xiaoshi Capsule (BXC) has a reliable curative effect on hepatolithiasis. The objective of the task is to research the mechanism of BXC.Feeding guinea pigs with lithogenous diet with low protein, BXC was administrated for 3 months at the same time. In the end, the gallstones formation rate, biochemical indies and hepatic morphology were observed. Furthermore, the bacteriostatic effect in vitro, the effect of excreting bile, the spasmolytic effect in vitro, anti-inflammatory effect and antianalgesic effect of BXC were also researched.The results showed that pigment gallstones were found in the gallbladder of guinea pigs, and the gallstones formation rate was 76.92% in model group. The lithogenous diet with low protein led to the hepatic morphologic injury firstly, such as the fatty change of liver, the enhancement of the activity of endogenous p-Glucuronidase, the total ATPase and Ca2+Mg2+-ATPase. The metabolism of bile acids was abnormal, such as the decrease of the concentration of glycocholic acid in bile. In addition, the concentration of direct reacting bilirubin in bile decreased. At the same time, the percentage of indirect reacting bilirubin and the concentration of Ca2+ and H+ increased. All of these could accelerate the formation of pigment gallstones.Preventive administrating BXC, it could restrain the pigment gallstones formation significantly, and the gallstones formation rate was cut as to 20%—40.00%. The effect and mechanism of BXC on preventing the lithogensis of pigment gallstones in guinea pigs were concluded as follows: Firstly, BXC could weaken the activity of endogenous β-Glucuronidase in liver, and then decrease the concentration of indirect reacting dilirubin in bile. It could also decrease the concentration of Ca2+ in bile. |
