Psoralen Treatment Of Osteoporosis, The Substance Of Basic Research

Fructus Psoraleae is the dry and mature fruits of the leguminous plant Psoralea Corylifolia L. recorded in the china pharmacopoeia (2005 edition). Fructus Psoraleae is a common tonic Chinese medicine, which is pungent and bitter in flavor, warm in nature, and attributive to the kidney and spleen meridians; it invigorates the kidney and strengthens yang, arrests nocturnal seminal emission and reduces urination, warm the spleen and stop diarrhea. The doctors in past dynasties used Fructus Psoraleae for invigorating the kidney and strengthening the bones; the circs of clinical applications and modern study of pharmacology suggest that Fructus Psoraleae can assuredly strengthen the bones and cure osteoporosis, so it possesses well foreground of exploitation. The thesis is to screen out the active fractions and components from Fructus Psoraleae by experiment in vivo and in vitro. Basing these researches, we studied the method of determination of the total effective compounds for curing osteoporosis in Fructus Psoraleae, and the fingerprint of the effect fraction.Five fractions were got from Fructus Psoraleae through the systemic solvent distillation method, then screen out the active fraction by treating the osteoblast in vitro and the experiment of estrogen action in rat's vivo. The results indicated that given the high dose (5g/kg) of the petroleum fraction shows estrogen style function evidently (P<0.01) compared with the blank group; given the middle dose of the ethyl acetate fraction shows it can accelerate the proliferation of the osteoblast and enhance the activity,of ALP evidently, besides the activity is stronger than the masculine comparison group qianggu capsule.The petroleum fraction was separated by silica gel chromatography and compound I which have evident estrogen function was found by the experiment of estrogen action in rat's vivo. Compound I was identified to be Bakuchiol.The ethyl acetate fraction was separated by silica gel chromatography, then isolated and purified by silica gel and reverse silica gel chromatography, nine compounds were isolated and identified as Psoralen Isopsoralen Psoralidin...
