Tegafur is a kind of common chemotherapeutics to cancer which is widely used for treating malicious carcinoma. But the limited of its clinical use lie in its obvious toxicity and adverse reaction, complication and resistance to use long term drug. Enormous obstacles of chemotherapy is multiplex drugs resistance (MDR) that susceptibility of tumor cells to chemotherapeutics is reduced. Toxicity and adverse reaction of chemotherapeutics became increasing worse as its dosage was increased. To resolve the question, as one of important studying ways, low toxic and effective reverse agent of MDR was tried to research. The study showed that Fructus Bruceae oil is a kind of herb which has anticancer activity and can enhance immunity of body and it is an excellent DMR reverse agent which can inhibit growth of MDR tumor cells and enhance susceptibility of tumor cells to chemotherapeutics. The intention which combination Tegafur sub-micronized emulsion had been prepared with Fructus Bruceae oil and Tegafur was that susceptibility of tumor cells to chemotherapeutics and body's immunity would be enhanced and its toxic reaction would be reduced.Qualitative and quantitative analysis Measurement of Tegafur and Fructus Bruceae in combination tigufar sub-micronized emulsion was founded which content of Tegafur and its decompounded production was determined by UV and HPLC and Fructus Bruceae oil was determined by acid-alkai titration measurement and TLC. The method could control effectively quality of agent.The emulsification of single emulsifier, physical mixed emulsifier and phospholipids membrane emulsifier were evaluated by the method of surface (interfacial) tension isothermal curve comparison. Moreover, it was also verified by the Ke value and particle size distribution of Fructus Bruceae oil emulsion.The effects of Fructus Bruceae oil, emulsifier, buffer solution on the emulsion were studied by single factor test with method of appearance, particle size and distribution, centrifugalization and KE as investigation index. Effects of emulsification temperature, ultrasonic intensity and time, nanomizer working pressure, cycle times and emulsification time, pH and sterilization were also investigated by single factor test and orthogonal design.The physicochemical properties of combination Tegafur sub-micronized emulsion were studied. The microphotograph showed that most of emulsion drops were round in shape.
