Part I : The culture and the monitor on immunocytochemistry ofneonatal rat cardiac fibroblastsObjective Monitor the marker of the neonatal rat cardiac fibroblasts to prepare for the nest researches. Methods To observe the expression of Vimentin,α -Smooth Muscle Actin , desmin and Von Willebrand Factor in the cell that we culture by in vitro culture and immunocytochemistry staining. Results The staining of monoclonal antibodies against Vimentin were positive, while the other staining were negative for the antibodies against α -Smooth Muscle Actin and Von Willebrand Factor. Conclusion: what we cultured were the cardiac fibroblasts.Part II: Effects of tetramethylpyrazine on the production ofNO, TNF- α and TGFβ1 in cardiac fibroblasts in neonate ratObjective To investigate the effects of tetramethylpyrazine on the production of endothelin(ET) , nitrogen monoxidum (NO) , tumor necrosis factor α (TNF-α ) and transforming growth factor β1i(TGF β1) in cardiac fibroblasts (CFs) in neonate rat. Methods CFs were isolated by trypsin digestion method, the changes of ET, TNF- α , TGF β1 and NO production from CFs were examined by radio immunoassay, nitrate reductase-dependent assay and enzyme linked immunosorbent assay(ELISA) respectively. Results TMP(5mg/L-20mg/L)decreased ET, TNF- α , TGFβ1 synthesis in a concentration-dependent manner(P<0.05), but showed no significant changes to NO production(P>0.05). Conclusion These results showed that TMP can decrease the production of ET and influence the balance of these factors. TMP may inhibit myocardial fibrosis by modulating these cytokines.
