Objective:In order to investigate the alteration of β-amyloid precursor protein (β-app) in neurons of Tat brain penumbra following traumatic brain injury and evaluate neuroprotecting effects of magnesium. Methods:Fifty SD rats were randomly assigned into five groups: the control, the injury and three treatments. Rat traumatic brain injury was established according to feeny's method. The treatment groups was treated with three levels usages of MgSo4 (35mg/kg 日,100mg/kg. 日, 300mg/kg.日) .Animals were killed at 3 hours, 12hours, 1day, 4days, 7days after trauma. β-app was detected by immunohistochemistry. Result:1)Compared with the contral group, the immunoreactivity of β-app began to increase significantly in the penumbra at 3 hours after rat traumatic brain injury.There was twice increase at 12 hours and in 4 days: The imraunoreactivity of β-app reached the lowest level of increase in 1 day.But,it still had significantly difference compared with the contral group.After 4 days, the immunoreactivity of β-app began to decreae in the penumbra.2) Compared with the injury group, treatment group could restrain expression of β-app. As soon as possible using magnesium would prevent sharp change of espression of β-app that was caused by developing injury of nerve cell.
