| Major Histocompitibility Complex (MHC/H-2) is the very important gene cluster to decide the immunogenetics of mouse. Especially, the H-2D and H-2K are the most reactive and critical gene sequences of this cluster. Therefore, by carrying out the Genetic Monitoring of H-2 gene in Inbred Strain of Mice, it can control the efficiency and quality of immunogenetics of mouse. Besides, it can improve the evaluation technology of experimental animal.There are totally three parts in this paper. In parts one, firstly, Microcytotoxicity test was set up. And, the international standard monoclonal antibodies of Major Histocompitibility Complex were introduced, such as H-2D~b(27-11-13), H-2D~d(34-5-8), H-2D~k(15-5-5.3), H-2K~k(16-3.22.4). Meanwhile, the domestic standard monoclonal antibodies were introduced as well and they are H-2D~b(2), H-2D~d(4), H-2D~k(32), H-2K~b(33), H-2K~d(31), H-2K~k(23). By adding special foreign monoclonal antibodies in the spleen cells of the inbred strain mouse, the results showed that antigen and antibodies reacted intensely with each other, resulting in the death of the same modal spleen cells. By observing the percentage of the dead cells over the total cells under microscope, various strains of H-2 genes can be determined. In addition, by comparing and analyzing the international and domestic monoclonal antibodies, the results indicated that there are no distinct differences between them.In part two, based on the DNA sequences of H-2D and H-2K genes,... |
