The chronic accumulational arsenic poisoning is common. The morbility is very hiding and the course of disease is long. But the traditional medicine has some predominances in treatment. In past years,some clinical and empirical studies displayed that the traditional medicine have peculiarity in treatment. This experimental purpose to study and research Chinese crude drug therapeutic pharmacodynamic action and mechanism of action. The experiment also provid experiential for clinical application.Antidotarium is my tutor's empirical formula,that is summarized from his clinical experiences and theory investigation. The antidotarium have some effectiveness such as elimination of toxicant,deintoxication,accommodation and recruit. It is established to aim directly at the pathogenesis of the arseniasis that is turbid poison in body,stagnated blood stay inside and healthy energy to be injured. It include green gram, licorice root, coptis chinensis, beef-steak plant leaf, zingiberis recens, paeoniae radix, lalang grass rhizome, prepared pieplant,chuling. The antidotarium be used in therapy for arseniasis,that curative effect is very good. The design of experiment is to set up a animal model for chronic accumulated arseniasis.,Then to give Chinese crude drug intervention. From it we can investigate pharmacodynamic action and mechanism of action of the antidotarium.In (2000), realgar essential component is arsenic disulfide,and contain a little arsenic trioxide and some microelement. The content of arsenic is exceed ninety percent. The arsenic trioxide's quantity to determine the toxicity of realgar. This experiment use realgar intragastric administration once every day that can cause Jimpy mice arsenic poisoning. According to the preliminary experiment and literature record,the dosage of realgar to be choosed is 2mg/kg in 9 weeks. The experiment's grouping is normal control,five week model group,nine week model group,westerm medicine control group, Chinese crude drug middle dose group and Chinese crude drug large dose group. According to the contents of arsenic in blood and urine,to discuss the role of discharge arsenic about antidotarium; determination of ALT,Cr contents and detection of liver and kidney pathology to study realgar led to jimpy mice's liver and kidney damaged; determination of GSH-Px,SOD to study the role of antidotarium affecting the cellular oxidation. In a word,basis on the index we can investigate the toxic mechanism of action about realgar and reaserch the deintoxication of antidotarium.The result display that: the realgar not have acute toxicity. It led to a chronic accumulational arsenic poisoning but toxicity is very small. This experiment show that realgar is not easy to cause liver and kidney damaged. If we normative to use realgar in clinic,the damage of health is not happen. It is successful to use realgar founding arseniasis model,but if the dose and cycle are not same the degree of model also different. So the standard of successful arseniasis model need to the more deeping discussion. The experiment confirm that the realgar poisoned because realgar include an element that is arsenic,so it have cytotoxicity that is to influence celled antioxidation ability particular the enzyme including sulfhydry. Antidotarium have the reducing arsenic in blood and urine,it means the antidotarium have eliminating arsenic role specially in urine but in bood. And it can depress the excitable heighten of the GSH-Px and SOD that explain the antidotarium have regulation roles of celled antioxidation ability that led to deintoxication. Compared to western medicine ,the chinese crude drug have the same curative effect and deintoxication, also dose have not direct ratio relation, but pathology of liver and kidney display the traditional chinese medicine has protection function and the traditional chinese medicine cruative effect is better than western medicine. In this experiment, realgar caused damnification for liver and kidney is very little,so can not reflect the pharmacodynamic action of Antidotarium ,that maybe relate with design of experiment. Also it is disadvantage in the experiment,so it need further study and research.
