The ointment extracted from Plastrum Testudinis with modern technologies andtraditional technologies was studied. Ointment extracted from Plastrum Testudiniswas separated by silica gel column chromatography, petroleum ether-ethyl acetatewas used as the eluant, sixteen components were obtained. To observe the exactchemical compounds GC-MS was carried out. In order to screen out the effectivecomponents MTT experiments on the proliferation of mesenchymal stem cell (MSC)were also executed. Results of the activity indicated that cell viability in components2, 3, 11, 12, 16 with 0.825g/L, 2.475g/L and4.95g/L concentration groups wasincreased than that in control group (p<0.05), cell viability in component 4 with0.825g/L,2.475g/L and4.95g/L concentration groups was decreased than that incontrol group. Cell viability in the other components was similar to that in controlgroup (P>0.05).The compounds which play main functions were forecasted based on the resultsby GC-MS and the activity datas on rMSCs. Also these compounds were validatedby HPLC and nine standard compounds. The Ts-12 and Ts-4 was choosed as theexamples in HPLC experiment. Hexadecanoic acid, methyl ester (S-1),hexadecanoic acid, ethyl ester (S-3), octadecanoic acid, methyl ester (S-4),Cholesterol (S-7), and tetradecanoic acid Cholesterol ester (S-8) were the maincompounds of Ts-12. Octadecanoic acid (S-5) was the main compound of Ts-4. TheMTT assays and flow cytometry were carried out in order to study the activities onrMSCs of nine standard compounds. Results of the activity indicated that cellviability in tetradecanoic acid Cholesterol ester (S-8) and hexadecanoic acid, methylester (S-1) groups was increased than that in control group (p<0.05), cell viability inoctadecanoic acid (S-5) groups was decreased than that in control group.To Hunt the homologies of Chinese traditional herbs which have the similareffects on rMSCs the technology of fingerprint was used. The fingerprint of volatileoil of Plastrum Testudinis based on the results of GC-MS and MTT. Compared extracts of SWT and extracts of the seeds from the wild Calophyllum withfingerprint of Plastrum Testudinis. The validity of comparison other Chinesetraditional herbs with fingerprint of Plastrum Testudinis was 86%. So the fingerprintbased on the activity was scientific, and this method provided a new way to studythe herbs.
