Objective: To discuss the methods of separatiing, purifying and fostering cardiac muscle cells and their ability of mesenchyma stem cells under micro-environment of different cardiac muscle cells.Methods: To separate and culture the new-born mice's cardiac muscle cells by differential and adherent culture. This experiment is devided into three groups: the contact and joint culture group which plants cardiac muscle cells and MSCs by the ratio of 2:1 in the same culture dish; the non-contact and joint cultrue group which cultures the coverglass with the cardiac muscle cells and BMSCs planted by the ratio of 2:1 in a culture dish; the contrast group which cultures BMSCs in routine method. After a week to examine the cTnT expressions of the three groups.Result: In cardiac muscle cells culture, there is statistic expression between the differential and adherent 60min group and the 90min group. There is statistic expression between the first two groups and the group of 120min. (1) The cells cultured by the contact and adherent culture group expresses cTnT; (2) The cells cultured by non-contact and adherent group expresses cTnT; (3) The cells cultured by the contrast group does not express cTnT.Conclusion: In the course of cardiac muscle cells culture, the number of cardiac muscle cells cultured by differential 60min is much higher than differential 90min and 120min. Both contact group and non-contact group can make BMSCs induce and devide into cardiac muscle type cells but the contrast group can not.
