OBJECTIVE: To discuss the protective effect to the rats sufferingacute pancreatitis by using Dahuang and Dahuang compound. METHODS:Fifty male Wistar rats were randomly divided into 5 groups: Group1: Sham the rats as a control: Group2: Adopted improved method"retrogradebile common duct injection with Sodium taurocholateindirectly through duodenum", Acute pancreatitis model wereestablished by retrograde pancreatic duct injection with 3% Sodiumtaurocholate at the dosage of 1ml/kg weight. When the operation werecompleted, the whole rats were all injected subcutaneously with 0.9% NaCl 5 ml on the back; Group 3, 4 and 5: The model were establishedin the same way like group 2. 2,10 and 18hours later, The rats wereinjected from lavage-needlepoint respectively with differentdecoction at the dosage of 0.9ml/100g weight: Dahuang,Xiaochengqitangand Taohechengqitang. Second, observed ordinarysituation of the rats in 5 groups and killed half of them 12 hourslater and leavings 24 hours later. Took blood sample formeasurement amylase, IL-1 and TNF-α. Incised pancreas and ileumfor pathologic examination. RESULTS: 12 hour later, it showedhyperaemia and edema on pancreases of the rats in group 2. Pathologicexamination showed that distance of pancreatic leaves and leafletsadded, and large amount of neutrophilic granulocytes infiltrated in the pancreas tissue. Evident edema, hemorrhage and parts of necrosisin the pancreas could be seen, and normal structure disappeared.Edema and necrosis in the pancreas worse increasingly with time.Pancreas of group 3 to 5 showed a little difference in each periodof time. The distance of pancreatic leaves and leafletsadded lightly.A few neutrophilic granulocytes infiltrated in the pancreas tissue.Parts of necrosis or edema only could be seen in the pancreas. Thewhole structure of leaves was normal. There was a little hemorrhagein the pancreas, pathologic changes 24 hours later were much gentlethan that 12 hours later. After 12 hours, mucous membrane of ileumdamage grade of group 2 was 3.8±0.84, AMYL (3401.8±2634.37),IL-1 (82.2±14.39), TNF-α(159.8±56.55), 24 hours damage grade(4.2±0.84), AMYL (9685.4±286.57), IL-1 (151±40.76), TNF-α(428.8±215.29); The same index in group 3: 12h (2.8±0.45),AMYL (2119.2±1260.83), IL-1 (59.8±12.07), TNF-α(103±17.22),24h (1.8±0.45), AMYL (1634.6±366.24), IL-1 (65.6±9.56), TNF-α(112.8±31.88); The same index in group 4: 12h (2.4±0.55),AMYL (1593±260.93), IL-1 (49.2±7.79), TNF-α(89±9.98), 24h(1.6±0.55), AMYL (1368.6±270.99), IL-1 (61.8±4.15), TNF-α(106.4±16.96); The same index in group 5: 12h (2.4±0.55),AMYL (1130±93.86), IL-1 (29.6±3.36), TNF-α(84±4.47), 24h(1.2±0.45), AMYL (820.8±40.23), IL-1 (35.2±10.5), TNF-α(68.6±13.13). The difference was significant. Conclusions: Dahuang and all the Dahuang recipes are effective to the rats ontreating acute pancreatitis. The effect of Taohechengqidecoction is the best.
