Recently, much attention are paid to the interaction of small molecule drug with DNA.Chlorpromazine hydrochloride and Promethazine hydrochloride are two commonanti-psychotropic medicine, belonging to phenothiazine derivative. The structure andelectrochemical character of Chlorpromazine hydrochloride and Promethazinehydrochloride in the solution were studied in this paper.The complete assignment of 1H and 13C resonances from Chlorpromazine hydrochloride inchloroform has been done by NMR in this paper. In virtue of mass spectrometry, therupture mechanism has been primarily concluded.According to spectroscopic and electrochemical methods, the interactions with DNA wereinvestigated. And the interactive modes were analysed with the information of structure insolution. The UV-spectrum illustrated intercalative binding existed in both two druggs, andthe interactions with dsDNA are stronger than ssDNA.The results of electrochemical experiment illustrated as follows. Chlorpromazinehydrochloride showed electrostatic and intercalative mixed bindings with DNA in solution.Promethazine hydrochloride mainly showed intercalative binding with DNA. With electrontransfer dynamics, the electrochemical parameters at the electrode surface werecalcaulated.Finally, the characters of coordination and as a surfactant were studied. The surfactant ofdrug need much more work to do in future.
