Interaction between Trypsin and CamptothecinThe interaction of trypsin with camptothecin(CPT) in vitro was studied by ultraviolet(UV) absorption spectral and fluorescence spectral methods. Making out value of Ki (4.36×10-3mol·L-1) according to the ratio between 1/v and the amount of inhibitor contributes to the conclusion that CPT is a noncompetitive inhibitor. The interaction between CPT and trypsin is quite strong.CPT can affect the conformation of trypsin in sone degree. Fluorescence quenching contributes to nonradiative energy-transfer, which results a static quenching of CPT to trypsin. Their binding constants and the binding sites of CPT were determined. ARPE-19 is an adult retina pigment epithelial cell and we have established the oxidative damage model of ARPE-19 cells which were dealted with acrolein. Mitochondrial nutrients is a kind of intermediate metabolite or exogenous small molecules, maintaining mitochondrial function, clearing the electron transport chain helping reduce oxidative damage and protecting cells from damage. In this study, we have used the retinal pigment epithelial cells of acrolein damage model to research the protective of mitochondria nutrients. Research shows lipoic acid can protect acrolein-induced mitochondrial damage. One of the cauuses of Parkinson ' s Disease (PD) is the enchancement of oxidative stress, mitochondrial dysfunction as well as the function protein deposition, and which finally causes the lose of dopaminergic neurons in mesencephalon. Therefore, improvement of mitochondrial function, elimination oxidative damage in organism and suppression or delay of the function protein deposition are the nessessential methods and measures of preventing PD; B vitamins group is a vital role kind of trace element in the energy metabolism process, some of which or simply modified product are the compoundauxiliary bases of mitochondrial electron transfer chains; The sufficient B vitamins group supply is one of the effective ways to dredge the electron transfer chain, repair the multiple mitochondrial damage and eliminate free radical.At present prevention function of B vitamins group for PD is still not very clear. We use the the PD model of which MPP inducts the cells to research vitamins' prevention function to PD. Finally, it was indicated that vitamin B3 can obviously delay mitochondrial oxiditive damage, raise mitochondrial enzyme activity and protect mitochondrial,which indicates vitamin B3 has certain prevention function to PD.
