Traditional Chinese medicine(TCM)compound is the main form of medication of Chinese medicine.The theory of compound compatibility is the essence in traditional Chinese medicine compound.Matched pair of medicine(MPM)is a special from of compound compatibility and is the core of prescription.Researching matched pair of medicine has important significance for analyzing compatibility law of herbal prescription and developing new drugs.This paper studies the chemical constituents and pharmacokinetics of matched pair of Radix Polygalae(RP)end Rhizoma Acori Talarinowii(RAT)by the methods of GC-MS and HPLC.This research is hopeful to elucidate the mechanism in vivo of the matched pair of RP and RAT,to provide reference for researching compound compatibility theory and new approaches for developing innovative drugs.This thesis consists of five chapters,and author's main contributions are as follows:1.The liposoluble components and volatile oil of RP,RAT and matched pair of them were identified by GC-MS.The results indicated that the compatibility of above pair had obvious effect on volatile oil and liposoluble components.2.Established the HPLC analysis method for RP and studied the changes of chemical constituents in RP after compatibility with RAT.The result showed that chromatogram peaks were summated,peak area increased and two new peaks were found.It suggested that the composition dissolution of RP was increased with the addition of RAT,chemical changes may be occurred after compatibility. Established the HPLC analysis method forα-asarone in RAT and researched the content change ofα-asarone after compatibility.The content ofα-asarone decreased after compathility of two medicines,it showed that the adding of RP inhibited dissolution ofα-asarone.3.Established the HPLC analysis method of 3,4,5-trimethoxycinnamylic acid (TMCA)in plasma with the index composition TMCA which was metabolic product of RP,and this method was evaluated in pharmacokinetics research of matched pair of RP and RAT.The results indicated that the pharmacokinetic parameters of TMCA conformed to the two-compartment open model pre-and post compatibility.Cmax,AUC0-∞increased significantly,CL/F decreased after being compatibilitied,suggested.that compatibility makes the bioavailability of TMCA improved and slow down the elimination.Also the effect lasted for a long time.
