Liuchuanyu (LCY) is the dried whole plant of Linaria vulgaris ssp. Sinensis (Bebeaux) Hong, which has effect on alleviating fever and disintoxication, dissipating blood stasis and promoting the subsidence of swelling. Flavonoids are its main components, modern pharmacological research shows that it has good effect on cardiovascular system. To provide the basis for the effective application of this plant, we made research on the quality control of Linaria vulgaris ssp. sinensis (Bebeaux) Hong, and the preparation technology of its extract. The following studies were designed.Literature advances A summary of the chemical constituents, pharmacological effect, clinical effect and quality assessment of linaria species.Research on chemical reference substance of LCY Four compounds were separated from Linaria vulgaris ssp. Sinensis (Bebeaux) Hong with solvent extraction and column chromatograph such as macroporous adsorption resin, polyamide and Sephadex LH-20 chromatography, and the compounds were identifed as hesperidin (1), pectolinarin (2), acacetin (3) and pectolinarigenin (4). Among them, hesperidin was separated from this genus for the first time. Analyzed by HPLC, their purities were 98.5%,99.2%,99.2% and 99.0%, respectively. They meet the requirement for reference substances on qualification and quantification analyses. Research on quality control method of LCY TLC and HPLC methods were carried out to identify Linaria vulgaris ssp.sinensis (Bebeaux) Hong for the first time. In TLC, Pectolinarin was used as reference substance, and in HPLC, four peaks were identified as hesperidin, pectolinarin, acacetin and pectolinarigenin, respectively. Water content, total ashes and ethanol-soluble extract were determined. We found that the water content and total ashes in different parts of raw material collected from different year have no big differences. However, the condition of ethanol-extract are not the same, it has high content in flower, and then in fruit, leaf and stem, respectively. HPLC method was established for simultaneous determination of four compounds including hesperidin, pectolinarin acacetin and pectolinarigenin with linear ranges in 50.25-502.5μg/mL (r=0.9991), 30.68-306.8(r=0.9994), 1.98-19.8μg/ml (r=0.9990) and 2.03-20.3μg/ml (r=0.9994), respectively, with the average recoveries 99.1-100.7% (RSD 2.1-3.3 %), which shows good accuracy, reproductivity, and repeatability. Research on the preparation technology of LCY-extract Take total flavonoids as indexes, orthogonal experiment was carried out to optimize the extract factors as: C2 multiple of D3% ethanol extract for B3 times, A1 min every time. The purification factors with macroporous resin of LCY were also investigated. The result shows that total flavonoids can reach to 56.8% of LCY-extract after purification technology.
