Recently, the incidence of diabetes mellitus (DM) was increased sharply,sowas the diabetic nephropathy (DN) as the complication of DM. The pathogenesisof DN was complicated, which called the attention of researchers in the renalfield. The most important factors in the pathogenesis of DN were the heredity andthe hyperglycemia, and the cytokines might play important poles in the process ofthe disease. Of all the cytokines, many resear chers pay the attention to theVasular Endothelial growth Factor (VEGF)."Haikunshenxi"was the nationalâ…ˇgrade new Chinese medicine to treatCRF and early-stage uremia. It was compos ed by Fucoidan (FPS). The researchesshowed that FPS had many functions of biology, such as stepping the bloodpressure, blood fat and blood sugar down, enhancing immunity and so on. Toelucidate the mechanism of action of Haikunshenxi preventing and curing DN, weresearched it from levels of or gan, tissue, cell and protein by animal experimentand vitro cell experiment.ObjectiveIn order to elucidate the mechanism of Haikunshenxi preventing and curingDN, we observed the effectivity of Haikunshenxi by established DN rats'model.We also research ed the effection of Haikunshenxi to VEGF from levels of or gan,tissue, cell and protein .Method1.Male SD rats were randomly divided according to weights. Then modelgroups were injected streptozocin (STZ) to induce DM model. Each treatmentgroup was treated with homologous medicine. Rats'weight was measured weekly,and 24h Urine protein was measured every four weeks. The kidney was taken atthe end of the 12th week to be observed the changes of the pathomorphology, andbe estimate d the pathology change s by glomerular sclerosis index numbe r and tubulirenales -mesenchymal injure score, at the same time the blood serum was separated to examinating blood biochemical indicator.2. The VEGF protein was detected in rats'renal tissue by immunohistochemicalmethod.3. With high glucose stimulating GMCs, the proliferation was detected byMTT method at 12h, 24h , 48h and 60h.4. With high glucose stimulating GMCs, the VEGF proteic content wasdetected by western blot method .ResultResults1. After the Haikunshenxi treatment, the weight of rats rised conspicuously,24h Urine protein reduced. BUN, Scr, TG and the hypertrophy of the rats kidneydropped obviously. The changes of pathomorphology of rats kidney wasameliorated obviously.2. Haikunshenxi declined the expression of the VEGF protein in cytoplasm ofrenal inherent cells of DN rats.3. Haikunshenxi suppressed the multiplication of GMCs stimulated by highglucose ; the large dose suppression effect was the best.4. Haikunshenxi suppressed GMCs to secrete the VEGF protein obviously,and showed the dosage dependence relations.Conclusion1. Haikunshenxi improved the kidney function of DN rats, reduced the urineprotein, reduced the pathology damage of kidney, and showed the dosagedependence relations.2. The mechanism of Haikunshenxi prevented and cured DN . In cellular level ,Haikunshenxi suppressed the multiplication of GMC s, which was possibly one ofthe mechanisms that Haikunshenxi prevented and cured glomerular sclerosis. Inthe protein level, Haikunshenxi declined the VEGF protein expression in kidneyorganization and GMCs then decreased the risk of the kidney fibrosis , and thatsuppressed the multiplication of GMCs, and reduce d ECM, thus delayed DN.
