Objectiv e:Nerve regeneration chamber of sciatic nerve injury model was set up,to investigate the effects of BMSCs transplantation on the regeneration and repair of function in sciatic nerve injury in rats.and to explore its mechanism for clinical treatment and provide a theoretical basis.Methods:BMSCs were isolated and cultured with complete bone marrow blood method.Trypsin and EDTA were used to digest and passage the cells.Spread to the third generation of mesenchymal stem cells using flow cytometry test surface anti-CD44,CD45, CD71 expression and thus on mass culture of mesenchymal stem cells identified.32 adult rats were divided into two equal teams at random.That is the experimental group and control group, each group 16.All of their left sciatic nerve were sharply transected and immediately repaired with 9/0 nylon sutures.Formation of nerve regeneration Room.BMSCs of passage 3 with 50ul culture fluid was pushed into distal stump of BMSCs treated group;50ul pure culture fluid was pushed into distal stump of control group.control group.Gross observation,electrophysiological examination and immuhistochemistry were carried out at 4weeks and 8 weeks post-operation.Results:BMSCs showed spindle-shape and fibroblastic morphology,forming a voflex arrangement,with uniforlll shape for P3 in culture.Under the light microscope.The special mark antigen of CD45 is negative,CD44,CD71 were detected and opsitive express by immunocytochemistry.In gross observation,there were slight ulcer on control team rabbits' treated feet.but nothing on the feet of MSCs treated team.Observating the distal stump with unaided eyes.there were muscle fibre aroud the nerve,after clearing the fibre,the good repaired distal stump can be saw.After the first four weeks and eight weeks of experimental rats sciatic nerve function index(SFI),nerve conduction velocity,Weight and gastrocnemius muscle fiber cross sectional area than the same period of the control group(p<0.05).After the first eight weeks of the inspection group found that immune mesenchymal stem cells into Schwann cells and experimental group Schwann cells than the control group increased significConclusion.BMSCs in vitro can be transplanted to the body,after transplantation in the local micro-damage to the environment through the Schwann cell transformatio n and the promotion of Schwann cell differentiation and proliferation,a nerve injury to the repair and regeneration. |
