Objective:To investigate the relationship of fibrinogen levels in plasma and the gene polymorphism with recurrent spontaneous abortion.Methods The 455G/A polymorphism and 148C/T polymorphism of Fg gene in 60 recurrent spontaneous abortion patients(case group) and 60 healthy non-pregnant women(control group) was detected by reaction-restriction fragment length polymerase chain polymorphism(PCR-RFLP);The plasma Fg level was detected by turbidimetry.x~2 analyses were used to compare Genotypic frequencies and allele frequencies between tow groups,and odds ratio(OR)was acquired to estimate in the same way.One-way ANOVA was used to compare Fg levels among the different periods of abortions.Independent-samples T test was used to compare Fg levels among groups,genotypes and the times of abortions.Result:(1) The genotypes and allele frequencies of Fg-455G/A and Fg-148C/T were not different between the case group and the control group.(2) Allele -455 G and -148 C or allele -455 A and -148 T of Fg-455G/A and Fg-148C/T in the case group and control group was close linkage disequilibrium, the rate reached 86 percent.(3) The fibrinogen levels in plasma were not different among the genotypes of Fg-455G/A and Fg-148C/T.(4) The plasma Fg in case was significantly lower than in control(t=-5.181,P<0.05).(5) The plasma level of Fg in the early abortion group was markedly lower than that in the late abortion group.(6) The plasma level of Fg in the more than twice abortion group was markedly lower than that in the twice group(t=4.308,P<0.05).Conclusion:(1) The low level of Fg is a probably risk factor of RSA.(2) The low level of Fg may be correlated to the early abortion and the number of abortions.(3) The polymorphism in Fg-455G/A and Fg-148C/T may be not corrected to recurrent spontaneous abortion.(4) The polymorphism in Fg-455G/A and Fg-148C/T may be not corrected to fibrinogen levels in plasma. (5) Allele -455 G and -148 C or allele -455 A and -148 T of Fg-455G/A and Fg-148C/T was close linkage disequilibrium.
