Objective:To investigate the effect of ganoderma lucidum spores on MFS,changes of HA contents , the expression of GFAP and activity of Glutamine Synthetase (GS ) in hippocampus of rat with epilepsy induved by PTZ. And further the study of epilepsy mechanism and the anti-epilepsy mechanism of ganoderma lucidum spores. Methods:36 male SD rats(weighing 180-220g ,purchased from Jiamusi university) were randomly divided into three groups: (1) the control group(n = 12), in which rats received intraperitoneal injections and oral administration of the normal saline and, (2)PTZ group (n = 12), in which rats were injected with a subconvulsant dose PTZ (35 mg/kg in 0.9% saline) intraperitoneally and received saline orally; (3) Ganoderma Lucidum spores group (n = 12)in which rats were injected intraperitoneally with PTZ (35 mg/kg in 0.9% saline) and received oral administration of Ganoderma Lucidum spores ( 300 mg/kg suspended in normal saline )。28days later withdrawal drugs for a week,then these rats received a final challenge dose of PTZ (35 mg/kg) to check the persistence of enhanced susceptibility to the convulsant. Only those PTZ-treated rats shows stage 2 convulsion for at least 5 times continuously were used in the study. Control rats received injections of saline instead of the PTZ challenge dose. At the end of the experiment ,four rats in each group were anesthetized with diethyl ether and perfused transcardially with 200 ml sodium sulfide medium followed by 200 ml 4% phosphate buffered paraformaldehyde for Timm's stain.The other 8 rats in each group were anesthetized with diethyl ether and decapitated , one of their hippocampus were made into homogenate for HA and GS detection ,the other hippocampus were fixed with neutral formalin for GFAP detection.Results:All the rats in ganoderma lucidum spores group and epilepsy model group achieved emblazed standard. Compared with epilepsy model group, the latent period of ganoderma lucidum spores group had obvious difference, but they had no obvious difference at during time. In supragranular the Timm staining scores of rats in PTZ model group were significantly higher than the that in control ( p<0.01)and Ganoderma Lucidum spores group ( p<0.05) .There was no significant difference between the control group and Ganoderma Lucidum spores group,The Timm scores in the CA3 region in rats of PTZ model group were significantly higher than those of control group(p<0.01) and Ganoderma Lucidum spores group(p<0.05) , and rats in Ganoderma Lucidum spores treat group have a higher Timm scores compared with control ones(P<0.05,) The resut of radioimmunity indicated that Hyaluronan increased signidicantly in PTZ model group(p<0.01,compared with control group), and they discreased after administration of Ganoderma Lucidum spores (p<0.01). Although it seems there were more Hyaluronan in Ganoderma Lucidum spores, but the diffrence had no significant。The astrocytes in Epilepsy group was significant more than that in control group ( p<0.05 ) and Ganoderma Lucidum Spores group(P<0.05),they also have more processes. Although astrocytes could be activated by seizure ,and their number increased ,the activity of glutamine synthetase(mainly expressed in astrocytes) which is the main synthetase in glutamic acid metabolism process decreased ( P<0.05, compaired with control group),and the activity of GS in Ganoderma Lucidum Spores group were higher than epilepsy(P<0.05)。Conclusion:Ganoderma Lucidum Spores could regulate the contents of HA and ease MFS, it also could regulate the function of astrocyte,and lighten seizures。...
