Objective To investigate the expression of Stomatin-like protein 2(SLP-2) in normal cervical tissue,precursor lesions and invasive cervical cancer and to analyze the correlation of SLP-2 expression with cervical carcinogenesis and prognosis.Methods Tissue microarrays(TMAs)(8×5) were used in this study to detect the expression of SLP-2 protein by immunohistochemistry.The correlation of SLP-2 expression with tumor differentiation,FIGO stage,recurrence and prognosis were evaluated.Results SLP-2 expression was significantly higher in normal cervical tissue(6.39±2.27) and lower in invasive cervical cancer(4.17±1.98) with precursor leisions(5.16±1.81) the middle.In invasive cervical cancer,SLP-2 expression was significantly lower in patients with their tumor maximum diameter greater than 40 mm(P=0.05) and deep stromal invasion(P=0.02).It was also significantly low-expression in patients with low grade differentiation(P=0.03),lymph node metastasis(P=0.03) and recurrence(P= 0.01).Multivariate analysis revealed that tumor size(P=0.001),SLP-2 expression(P= 0.011) and pelvic lymph node metastasis(P=0.016) were the independent factors that influenced cervical cancer recurrence.Both the DFS and OS in SLP-2 low-expression group were significantly lower than in SLP-2 high-expression group and pelvic lymph node metastasis(P=0.008) was the strongest independent prognostic factor for OS, followed by SLP-2 expression(P=0.010) and FIGO stage(P=0.032) by the Cox proportional hazards model for OS.Conclusion SLP-2 can be a novel cancer related gene and play an important role in cervical carcinogenesis.SLP-2 low-expressed in cervical cancer is associated with prognosis and can be a great valuable marker for cervical cancer prognosis. Objective To investigate the differential expression of Stomatin-like protein 2(SLP-2) before and after intraarterial chemotherapy(IAC) in local advanced cervical cancer(LACC) and the correlation of SLP-2 expression with therapeutic effect,prognosis and chemosesitivity was analized.Methods Tissue microarrays(TMAs)(8×5) were used in this study to detect the differential expression of SLP-2 protein in cancer tissues before and after IAC by immunohistochemistry.The correlation of SLP-2 discrepance with histological differentiation,FIGO stage and recurrence was evaluated and the relationship with therapeutic effect,prognosis and chemosesitivity was analysed.Results IHC study on TMAs showed that the expression of SLP-2 in cancer tissues after IAC was significantly higher than before IAC(4.33±1.97 vs.3.17±1.40,P=0.007), and both of them were obviously lower than in normal cervical epithelium(5.83±1.70,P<0.01).The discrepancy of SLP-2 before and after IAC was associated with tumor type (P<0.05),the depth of cervix stroma invasive(P=0.025),the histological differentiation(P=0.003),lymphatic metastasis(P=0.042) and relapse(P= 0.047).After IAC,the duration of follow-up ranged from 14 to 46 months(median,33 months) with 8 recurrences and 3 deaths.The recurrent rate was 33.33%.The mean disease free survival in SLP-2 up-regulated group was significantly longer than in down-regulated group(37.54 vs.17.40,P=0.0212).Conclusion SLP-2 protein is low-expressed in cervical cancer and up-regulated after IAC.The discrepancy of SLP-2 before and after IAC is associated with clinicopathologic features and prognosis,which demonstrates that SLP-2 can be a novel valuable marker for chemotherapy effect,prognosis and chemosesitivity.
