Fuzi (also known as aconite, monkshood) is the daughter root of Aconitum carmichaeli Debx. (Ranunculaceae), with the pharmacologic action of relieve pain, anti-inflammatory, antiarhythmia, increase interrenal system. Pharmaco research indicated that the main active components is alkaloid. Fuzi contains diester-diterpene alkaloids(C₈-Ac,C₁₄-benzoyl): aconitine(AC), hypaconitine(HAC), mesaconitine(MAC), toxicity of them are strong, 3-4mg can lead to death, in order to ensure the safty, Fuzi usually processed before using, the processing is just a long time decocting until the diester-diterpene alkaloids hydrolyzed.I researched the assaying meathod of alkaloid, having a comparing between acid-base titration and acid dye colorimetry, the consequence show acid-base titration which is more stable is better, I choosed it as the assaying meathod of total alkaloids of Fuzi. Establishing a meathod which determination the content of toxiferous component(diester-diterpene alkaloids) and effective component of Fuzi at the same time by HPLC.The purpose of this research is obtaining the teachnology parameter which abstraction and purifying alkaloid from Fuzi, the purity of alkaloid in the extractive is more than 70%, and formulating the quality criteria of medical material and extractive. Total alkaloids and benzoylaconitine as the evaluation index through the experiment, we get the teachnology parameter is:the powder of Fuzi, 0.1mol/LHCl water-solution (8 times of medicine material) extract 2 times, 2h/t, demesh the dopant of carbo, protein, tannin in extract by macroporous adsorptive resins, collect elutriant of 60% alcohol, hydrolyzed 2h in 120℃(0.2MP) adjusting the pH of digest to 10-11 with ammonia water, extracting 2 times(V/V:1/1) with acetic ether, recovering the acetic ether, drying it in low temperature.Semi-works production indicates this artwork is reasonable, stable, scientific, the purity of total alkaloids in extractive is more than 85%.
