Astragalus Root Prevention Of Oxaliplatin Chemotherapy Induced Neurotoxicity Of Basic And Clinical Research

The paper consists of three parts:a literature review,a basis of experimental and a clinical research.The literature review is about the advances in the treatment of neurotoxicity caused by Oxaliplatin in recent years.It includes clinical symptoms,mechanisms and the advances in management of Oxaliplatin-induced neurotoxicity with TCM and Western Medcine.Aggregate analysis,peripheral neuraotoxicity caused by Oxaliplatin is its main side effect and influence the patients'quality of life.According to the clinical observation,it proved that TCM is efficient in prevention and treatment of Oxaliplatin-induced neurotoxicity.The second part is a basis of experimental.Objective:To study the effects of astragalus root(a traditional Chinese herb medicine) on oxaliplatin-induced apoptosis of hippocampal neurons.Motheds:The primary culture of hippocampus neurons from SD rats(aged within 24h) were treated with different concentrations of oxaliplatin(L-OHP)(0,3,5 and 10μg/ml) and astragalus root(0.05,0.2 and 0.5g/ ml).The apoptosis of the neurons and the nerve growth factor(NGF) level were analyzed.Results:Astragalus root dose-dependently decreases neuronal cell apoptosis induced by oxaliplatin(P＜0.01) and increases NGF level(P＜0.05). Conclusion:Astragalus root can restrain hippocampus neuronal cell apoptosis induced by oxaliplatin,probably via increasing in NGF level.The third part is a clinical research.Objective:To investigate and measure the effects of the traditional Chinese medicine huang qi injection by transfusing in treating neuro-sensory toxicity by oxaliplatin treatment.Methods:There were 40 patients enrolled,among them the man patient's 23,the women patient's 17.A randomized controlled trial was performed.40 patients were divided randomly into A or B groups.Patients in B group was treated group,which treatd with chemotherapy combind with oxaliplatin plus the traditional Chinese medicine huang qi injection.A group was control group which treated chemotherapy alone.The more,the peripheral neuro-sensory toxicity was observed and measured nerve growth factor(NGF).Results:The reaction occurrenced amount of the nerve toxicity of the B group little in A group,and the degree was lightly,had only 6 patients'Ⅰclass to respond,had no ⅡⅢⅣclass to respond.(P＜0.01);NGF value A group decreased significantly in the second day(P＜0.01).Conclusion:the traditional Chinese medicine huang qi injection could prevent and treat peripheral neuro-sensory toxicity induced by chemotherapy combined with oxaliplatin.
