| OBJECTIVEPart ATo observe the relationship between proliferative diabetic retinopathy(PDR) and tumor growth factor-alpha(TNF-α),invest the proliferative diabetic retinopathy patient's serum levels of TNF-α,and provides an evidence to the Part B experiment.Part BTo research the influence of leech extract on cultured rhesus choroidal-retinal vascular endothelial(RF/6A) cells' secretion of TNF-αthat is induced by thrombin.METHODSPart A30 patients diagnosed with PDR were enrolled in the study along with 30 healthy persons(as the control).All people had serum TNF-αmeasured using ELISA tests.Part B①RF/6A cells were cultured in vitro with explants culture method.②RF/6A cells were treated with thrombin(20NIHU/ml) in different time(2, 4,6,8,10,12,24 hours).ELISA was used to evaluate TNF-αlevel.③RF/6A cells were treated with thrombin(20NIHU/ml),leech extract(64mg/ml), and thrombin associated with leech extract 10 hours.ELISA was used to evaluate TNF-αlevel.RESULTSPart Astatistically significant higher levels of TNF-αwere found in the group with PDR (137.92±4.14pg/ml) in comparison without healthy control group(50.04±1.75 pg/ml).Part B①Thrombin-induced TNF-αexpression exhibited unusual kinetics,reaching maximum levels after 2 to 10 hours,but decreasing to control after 10 hours. ②Thrombin-induced TNF-αexpression(0.07517±0.00207) is increased,when compared with normal control group(0.06017±0.00199)(P<0.05);leech extract group(0.03883±0.00302) compared with normal control group,TNF-αlevel decreased(P<0.05);thrombin associated with leech extract group(0.04467±0.00160) compared with Thrombin-induced group,TNF-αlevel decreased(P<0.05).CONCLUSION①Elevated levels of TNF-αin the serum occur in PDR,implicating a role for this cytokine in the pathogenesis of this retinal neovascularization disease-PDR.②leech extract down regulated the RF/6A cells expression of TNF-α.This experiment provides a new method to cure and prevent the retinal neovascularization disease... |
