The Notoginseng Diol Pretreatment Induced Endogenous Neuroprotective Effects Of Hsp70 And Tnf-alpha Mrna Expression In Cerebral Ischemia

Objects:To discuss and explore the relationship between Panasadiol Saponins preconditioning and endogenous neuroprotection by investigating the effect of Panasadiol Saponins preconditioning on the pathological change of brain tissue and the change of cerebral infarct volume after cerebral ischemia,as well as the expression of heat shock protein 70(HSP70) and tumor necrosis factor-α(TNF-α).Methods:SD rats randomly divided into six groups:blank,sham operated(SS+SS),model(NS+MCAO),ischemia preconditioning(IPC+MCAO),PDS preconditioning(PDS+MCAO),PDS treatment(MCAO+PDS ).Nonintevention to blank group;Model group was given gave 2h MCAO after 3-day continuously intraperitoneal injection of NS,and then executed after 22h reperfusion;IPC group was treated the same with model group except was given 10 min IPC before 3-day continuously intraperitoneal injection of NS;SS group was given 10min SS first and 2h SS after 3 days,executed 24h after;PDS+MCAO group was given PDS for 3 days before 2h MCAO,executed 24h after model building;MCAO+PDS group was given 2h MCAO and executed after 3-day PDS treatment.After model building,observation was focused on the loss of nerve function of rats,pathological change by HE dyeing, Nissl body by Nissl dyeing,infarct volume by TTC dyeing,HSP70 masculine expression by immunity tissue chemistry dyeing,and TNF-αmRNA express level by RT-PCR.Experimental data was dealt with SPSS13.0.Results:Panasadiol Saponins preconditioning can reduce the volume of cerebral infarction and protect neurone;It dramatically increases the expression of HSP70 and decreases the expression of TNF-αmRNA.The change above is similarly as strong as that of IPC group,but stronger than that of Panasadiol Saponins treating group.Conchlsions:Panasadiol Saponins preconditioning reduces pathological change,protects neurone,lowers the volume of cerebral infarction,increasse the expression of HSP70 and inhibits the expression of TNF-αmRNA。It is suggested in the experiment that the activation of HSP70 and the containment of inflammatory reaction may be the important mechanism of endogenous neuroprotection induced by Panasadiol Saponins preconditioning.
