Objective To compare the therapeutic effects of intravitreal injection and retrobulbar injection of Triamcinolone acetonide on treating uveitis.Methods Forty normal pigmented rabbits were divided into four groups randomly and were induced the experimental uveitis at the same time in the right eyes. The method of inducing the animal model is as follows: intravenous injection of 2% BSA 0.25ml as pre-immunization and intravitreal injection of 4% BSA 0.20ml as the second-immunization 7 days later. The animal model of uveitis are successful 24 hours later. Intravitreal injection of 4mg Triamcinolone acetonide and retrobulbar injection of 20mg respectively in per experimental group after the second-immunization 24 hours, the control group do the injection of physiologic saline in the same volume. The effect was measured by the grade of inflammation, cell count of aqueous humour,slit-lamp examination, fluorescein fundus angiography and histopathology on day 1,3,7,14 and 21 respectively and then analyze the data of experiment using statistical software;Results 1.The standard examination score of Uveitis demonstrates that Group A was higher than Group B(p<0.001)and there were no significant difference among sub-groups in Group A and Group B(p>0.05).2.Fluorescein fundus angiography examination: unclear fundus and failed FFA examination were seen on 7d in the control group; FFA shows papilla optica hyperfluorescence, retinal blood vessel circuity and leakage spots, capillary nonperfusion zone at 3 minutes 30seconds on 14d in the control group; FFA shows papilla optica normal fluorescence, no significant leakage spots at 1 minute 48 seconds on 7d in the experimental group of TA Intravitreal injection; FFA shows papilla optica normal fluorescence, no significant leakage spots and capillary nonperfusion zone at 1 minute 45 seconds on 7d in the experimental group of TA retrobulbar injection.3.Inflammatory cell countings of aqueous humour at 7 day: in Groups B1 and B2 were significantly less than Groups A1 and A2(P<0.05,P<0.05 and P<0.05,P=0.027); There were no significant differences between subgroups of each group(P=0.236 and P=0.372).4.Light microscope observation: Iris and ciliary body processes with inflammatory cell infiltrates were seen in the peripheral corneal stroma, limbus, and anterior chamber angle, Retinal cross section with inflammatory cells infiltrating the vitreous, retina, and choroid and no subretinal fluid was seen, but a detachment artifact was present on 7d in the control group. Iris and ciliary body processes with inflammatory cell infiltrates, Retinal cross section with superficial retinal inflammatory cell infiltrate but overall preserved architecture on 7d in the experimental group of TA Intravitreal injection. Iris and ciliary body processes with inflammatory cell infiltrates,Retinal cross section with almost normal architecture on 7d in the experimental group of TA retrobulbar injection.Conclusions It is successful to induce pigmental rabbit Uveitis using Albumin Bovine Serum ; It is worked using TA to treat Uveitis. Retrobulbar injection in TA groups has no difference with Intravitreal injection on treating uveitis.
