Adenovirus vector with plasmid of HK was infected into HCT116 cells,which were the negative control.And HCT116 cells were designed as the blank control group.In order to find out the changes of proliferation and invasion power of HCT116 cell lines,we analyzed the data by MTT,Boyden chamber and scratch experiment.RESULTS:The values of 490 nm wavelength light absorption were different in the three groups in MTT experiment.The one-factor analysis of variance show that the number of alive cells in the group of Ad-RhoAC was lower than others in the 3th-6th d(P<0.01).The repeated measure analysis of variance show that the cell proliferation was different in three group (F=67.772,P<0.001 ).The experiment of permeated artificial basal membrane showed that the invasion power of HCT116 cell lines are on the decline after treatment of Ad-RhoAC(32.67±6.66,55.33±9.87,63.00±9.64,P<0.05).The transwell experiment without artificial basal membrane showed that the immigration power of HCT116 cell lines are also on the decline after treatment of Ad-RhoAC(44.67±10.07,183.67±12.90,210.00±20.88,P<0.05).The scratch width of infected Ad-RhoAC group were wider than others.The cell pseudopod can also be seen in others.CONCLUSION:RhoAC siRNA gene mediated by adenovirus may be useful for inhibiting growth and invasion of HCT116.These results provide a novel therapeutic targetin preventing colon cancer cell invasion and metastasis. | | Keywords/Search Tags: | carcinoma of large intestine, siRNA, RhoA, RhoC, adenovirus | PDF Full Text Request | Related items |
