Objective:To investigate the role of IL-17 in acute rejection by detecting the dynamic change of serum IL-17(interleukin-17,IL-17) in the rat heart transplant rejection.Methods:We get healthy male Wistar rats with 45 and 15 SD rats in closed population.Experimental animal will be divided into two groups:SD-Wistar rats group are experimental group(n=15),while Wistar-Wistar rats being the control one(n=15).The modified heart transplantation is used to build the model.Getting the blood from the inferior vena cava on dayl,day3 and day8 post transplantation respectively.Monitoring the occurring of rejection daily is by touching the graft beating.Enzyme-linked immunosorbent assay (ELISA) is used to test serum IL-17 level in the transplantation model.The hearts was harvested and saved in 10%formalin,then embedded in paraffin.Results:The level of IL-17 in the experimental group in the first 1 day post transplantation was not higher than the control group(t=-2.080 p>0.05) However,the level of IL-17 in experimental group is higher than control group in the day3 and day8 group significantly(t=-3.126,-11.043 p<0.05 ). Histological examination shows the prolongation of post transplantation time results in severer of the extent of rejection pathological changes.Conclusions:The level ofIL-17 is raised in acute rejection of rat heart transplantation,and is positive correlated with its pathological class. Detecting the dynamic change of IL-17 may be can be a new target in early diagnosis of rejection in clinical.
