Objective To investigate the expression and significance of peroxisome proliferation-activated receptor-γ(PPARγ) in placenta with preeclampsia.Methods Placetal tissues were collected from normal pregnancies of the first trimester (first trimester group,n=18),the second trimester(second trimester group,n=12),the third trimester(third trimester group,n=15) and mild preeclampsia(MPE group,n=11),severe preeclampsia(SPE group,n=19).Immunohistochemistry was used to investigate the expression of PPARγprotein in placentaes of these women and the expression level of PPARγmRNA in the placentas were detected by semi-quantitative RT-PCR.Results(1) The expression of PPARγprotein was observed in the nucleus of cytotrophoblasts in the first trimester group and in the nucleus of sycytiotrophoblasts in the other groups.(2) The expression level of PPARγprotein in placenta of the first trimester group(0.389±0.006) and the second trimester group(0.390±0.003)were lower than the third trimester group(0.425±0.004,P<0.05).There were no significant difference between the first trimester group and the second trimester group.(3)The expression level of PPARγprotein in placenta of MPE group(0.291±0.005) and SPE group(0.213±0.008) were significantly lower than that in the third trimester group(0.425±0.004,P <0.01).The expression level of PPARγprotein of SPE group were lower compared with MPE group(P<0.05).(4) The expression of PPARγmRNA in placenta of the first trimester group(0.20±0.05) and the second trimester group(0.23±0.05) were lower than the third trimester group(0.43±0.06,P<0.05).There were no significant difference between the first trimester group and the second trimester group.(5)There were no significant difference of PPARγmRNA in placenta among the thrid trimester group,the MPE group(0.42±0.05) and the SPE group(0.39±0.12,P>0.05).(6)The expression level of PPARγprotein in placenta of preeclasim women with fetal growth retardation(FGR) (0.232±0.005)was significantly lower than those without FGR(0.272±0.005,P<0.05),but there was no significant difference of PPARγmRNA in placenta between them.(7) There was significant correlation between the level of PPARγprotein in placenta with preeclamsia and the quantity of 24 hour urine protein(r=0.517,P<0.01).Conclusion(1)The expression level of PPARγwas increased with the advancement of pregnancy.(2) The expression level of PPARγprotein in placenta of the preeclampsia group was lower than the third trimester group and correlated with the severity of the desease and the growth of fetus.
