Traditional Chinese Medicine (TCM) is the major form and method of clinical practice in China. It is a "black" system which includes numerous unknown compounds, and the multiple constituents are usually responsible for its therapeutic effects. The current quality standard of TCM is by measuring the concentration of one or very few markers or active components. Hence, this quality control standard could not reflect the real and comprehensive active constituents of TCM. Chromatographic fingerprint emphasizes on describing the integrity and synergistic effects of TCM, which just agrees with the characteristic of "black" system. Chromatographic fingerprint focuses on how to obtain more chemical information, thus the whole characteristic of TCM can be described better.Dangguibuxuetang (DBT) is a classical traditional Chinese formula widely used in clinic supplementing qi and blood. There are many chemical and pharmacological investigations performed on DBT. This thesis covers five aspects as follows:1. In the introduction part of this thesis, the recent developments of quality control, chromatographic fingerprints, modern analytical techniques with their application to TCM were summarized. Also, this part discussed the research status of Dangguibuxuetang.2. Fingerprints of DBT were analyzed using hydrophilic interaction chromatography (HILIC) and reversed-phase liquid chromatography (RP--LC). The retention behavior of weak polar compounds of DBT was analyzed in RP-LC mode. The peaks eluted in the beginning in RP-LC mode were still a complex system, the retention behavior of these highly polar compounds of DBT in hydrophilic interaction chromatography was presented.3. Relative adscription analysis of chemical components between Dangguibuxuetang and its single herb was conducted with the help of hyphenated instruments and related chemometric method. It was found that most components of the formal were from radix astragali and angelica sinensis. But these were changes in content. 4. The fingerprints of the petroleum ether, chloroform, ethl acetate, n-Butanol parts of DBT were constructed. Aso, this part found out the adscription of the every part peaks in Dangguibuxuetang and its single herb. It was helpful to state pharmacodynamics material foundation.5. The standardization of originaldata, examination of similarity degree and analysis of chemical pattern were achieved in Extendable Data-base for Quality Assessment of Traditional Chinese Herbal Medicine. The distinctions of the same batch of concentrated solution, extracta sicca and patent medicine of Dangguibuxuetang were found out. So it can provide a rapid method for the research complex medicines' quality control.
