The liverwort Marchantiaceae plants are well-known traditional Chinese medicinal herbs that are used extensively to treat skin tumefaction and protect the liver in rural zones in China. Most liverworts including Marchantia polymorpha L contain oil bodies which are mainly composed of mono-,sesqui-and diterpenoids and Iipophilic aromatic compounds. Lots of compounds from liverworts exhibit interesting organoleptic, pharmacological and other biological effects. So the phytochemistry of liverworts (Hepaticae) has been subject to intensive investigations over the last decades.In this paper, liverworts Merchantman polymorpha L collected in Mount of Hunan province was Phytochemically investigated, and the studies were performed on four ports as such follow:(1) The essential oils in Merchantman palwacea was extracted and analysed by GC-MS method. Adder to the amount of extracted essential oils, conventional hydrodisstilation extraction and Microwave-assisted extraction were investigated systematically. The microwave-assisted extraction method was better and the extraction process was optimized by the orthogonal design methed, with the yield of 3.51%. The compound in essential oils was Monoterpenoids and Sesquitepenoids.(2) Separation and Purification of the sesquterpenoids,steroids,Bibenzyls. Total crude extractant was separately absorbed and desorped well by column filled with silica gel, Aluminum oxide or carbon 18 material. According to the different concentration between before and after absorption, the effect of absorb-desorp by three column filling materials was compared. The results showed that silica gel is fit to separate and purify crude extractant. The process of separation and purification by silica gel was investigated systemtically. 10g/mL extraction solution was added into the column with the rate of 1BV/h up to silica gel being saturated absolutely. Total crude extractant was eluted by Petroleum Ether - Ethyl acetate with the rate of 1BV/h. Several compounds's purity detected by HPLC was all above 96%.(3) Separation and purification of flavones. The effect of Heat-reflux extraction and super voice wave-assited extraction on the yield of Total-flavones was compared. The factors influence process of super voice wave-assited extraction was analysised. The crude ethyl acetate extractant of Mt were separated by sephadex LH-20 and S-8 macroporous resin. According to the total flavones concentration, the two column filling material's effect in separating these flavones was compared. The HSCCC was used to separate and purified the crude total Flavone, the solvent System, flow rate and revolution rate's effect on separation process were investigated. The solvent composition of n-Hexene-ethyl acetate-ethanol-water is fit to separate crude flavone Compound, the purity of three flavone compouds detected by HPLC was more than 96%.(4) Structrual characteristion and HPLC analysis of seven compounds. NMR, MS, UV method was used to infer and identify the structure of separated compounds. According to the literature, the Structure of seven compounds was characterized.
