1 theoretical discussionLiver invading the spleen syndrome, a common type of depression patterns, is also known as "the wood overacting the earth". With good operability and repeatability, our group ensure the rat model subjected to chronic immobilization stress (CIS) was an ideal model for research on this syndrome. CIS simulates the pathological process of the liver failing to maintain the normal flow of QI and the treatment of Xiaoyao san promotes the functional recovery of liver governs smoothing qi flow(LGSQF). Since multiple systems relate to chronic stress, and different stressor, different individuals and different circumstances might lead to different results, it's so difficult for prevention and treatment to chronic stress diseases, whereas Chinese medicine, which with a concept of whole adjustment, treatment based on syndrome differentiation and individualized diagnosis, has shown greater superiority. Considerable interest in this paper has focused on the molecular mechanisms and regulation channel underly Xiaoyao san affording effective protection agaist this depression syndrome.Stress reaction is an adaptive response of all living organism to st ressful events and essential for their survival. In the stress response and development process, the three systems of the nervous, endocrine, immune are interrelated and mutually influence, the role of psychological stress take first place on the brain, which make the sympathetic-adrenal medulla system excited; While the most important way to change the endocrine function in the brain is that Hypothalamus- pituitary-adrenal (HPA) axis is activated. Under chronic stress, ctivation of HPA axis is the core of the pathological mechanisms, 'herefore, the focus of this issue of chronic stress on the HPA axis fter the adjustment on its.Experimental studyThis experiment is composed of two parts. Part 1, to establish an nimal model of depression induced by CIS in accord with syndrome of iver invading the spleen in TCM. From the animal behavior, observion he signs and symptoms and detection levels of serum D-fructose to onfirmate the rat model of syndrome of liver invading the spleen by IS; Part 2, the changes of HPA axis in the rat model of syndrome of iver invading the spleen, and Xiaoyao sans radioimmunoassay of regulation.By the ending of modeling the rats,we can observed the changes in HPA axis though the HPA axis indicators radioimmunoassay,and investigate the important regulatory role of the status Xiaoyao san.Part 1, the 54 rats were randomly divided into 3 groups:Normal, Model group, Xiaoyaosan group.3h daily bondage modeling for 21 days. The CIS rats were examined dynamically at the 1th, 7th,14th and 21st days by the following three aspects.①signs and symptoms, weight-gain, food intake water in take and levels of serum D-fructose;②open field test;③syndrome was evaluated by prevention and treatment of xiaoyaosan.Part 2, The above three groups of rats were sacrificed when 21 days of modeling. The rats were made of plasma, serum and hypothalamus homogenates for radioimmunoassay,the detection indicators were:Serum COR, plasma ACTH, plasma CRH and hypothalamic tissue CRH. Then observed the changes of HPA axis by the detection of these targets, and to explore the regulation of Xiaoyao san.3 results and conclusionPart 1:RESULTS:CIS group: The results revealed that subjected to CIS resulted in a decrease ofbehavioral activity, weight-gain, food intake, low levels of serum D-fructose, and weak ambulation, rising and decoration in the open field test especially during the period of 7th to 21st day. Xiaoyao san group: Xiaoyao san treatment significantly reversed the behavioral deficit of these depression model rats especially during the period of 14th to 21st day.CONCLUSION:With good operability and repeatability, the CIS rat model is an ideal model for research on syndrome of liver invading the spleen. Long term depression leads to liver invading the spleen, that is to say, results in syndrome of depression in combination with spleen deficiency. Xiaoyao san can regulate the behavior abnormality in CIS rats.Part 2:RESULTS:Model groups serum COR and plasma ACTH were significantly increased by the end of modeling ,while Plasma and hypothalamic CRH were significantly lower in the same time.There was no significant difference between Xiaoyao san group and normal group.CONCLUSION: By the end of modeling, HPA axis in the model group was on the state of that pituitary,adrenal cortex were hyperfunction, and the hypothalamic was secretion because of the negative feedback regulation. Xiaoyao san can significantly reduce the COR, ACTH levels,while significantly increase the CRH levels,so that can significantly regulate the change of the HPA axis.
