Micro-needle (Microneedle) transdermal delivery technology is the MEMS processing technology developed a new delivery method, the experiment for the first time Chinese herbal compound hydrogel Babu combined with the micro-needle technology to compound The Caulis as a model drug, through the optimization of formulation Cataplasm micro needle safety experiment, in vitro release experiments and animal pharmacokinetic experiments, the following conclusions:1. Selection of three extracts were prepared Sinomenium acutum extract,prepared by cataplasm pretest,comparing the characters. Cataplasm three methods influence the final choice by water extraction and alcohol precipitation method to prepare young acutum extract.2. To tack and hold adhesive force were observed, the single factor and orthogonal experiment, the initial Chinese Qing Bi Cataplasm optimized prescription, and then verified through repeated experiments and found that prescription of traditional Chinese medicine Qing Bi best Cataplasm ratio:5.30% of the section and the polyacrylate sodium NP-700, 0.36% glycine aluminum,27.70% glycerol,5.00% povidone K-90,0.36% of tartaric acid, 49.98% of distilled water,11.3% in capsaicin, borneol and sinomenium acutum extract.3. Micro-needle in the safety of the experiment:skin infection experiments showed that micro-needle into the skin does not promote bacterial infection in the body leading to infection. Micro-needle repaeatedly piercing the skin of vllunteers,security and stimulation experiments show that the micro skin against skin irritation small little effect,and the number of broken micro-needles in a safe range.Skin injury in the experiment, by comparing the micro-needles, acupuncture needles, and scraping the three methods, showed that skin injury for the smallest micro, high safety.4. Chinese Cataplasm paralysis by MEMS micro-needle Cataplasm in vitro release kinetic experiments to observe the cumulative drug penetration rate and the amout foud by using 200um micro-needles to penetrate 5N of force and the micro-needle skin pretreatment retention time of 3min when Transdermal best.5. Chinese Qing Bi Cataplasm microneedle transdermal delivery and processing of Chinese medicine Qing Bi Cataplasm after transdermal delivery of the pharmacokinetic study showed that:microneedles transdermal delivery after treatment Sinomenine AUC, Cmax increase large, can promote the absorption of sinomenine extend the elimination, metabolism slow down, to improve bioavailability.
