BackgroundAtheroselerosis is by far the most frequent cause of isehemic disease,the plaque disruption with superimposed thrombosis is the main cause of the unstable angina myocardial infaretion,stroke and sudden death. Previous studies showed that total flavonoids of Scutellaria barbata good antioxidants, the subject of the proposed antioxidant and anti-inflammatory from the angle barbata whether flavonoids inhibit the role of atherosclerosis.ObjectiveTo study the Effect and Mechanism of Total Flavone of Scutellaria Barbata on Atherosclerosis of ApoE-knockout Mice.MethodsFive weeks old APOE-/- mices were randomized into simvastatin positive control group [5mg/ (kg.d)] , The total Flavone of Scutellaria Barbata high dose group [200mg/ (kg.d)],Middle dose group [100mg/ (kg.d)],low dose group [50mg/ (kg.d)] and blank control group. At the end of eight week,all mice were killed.the serum levels of Triglyceride(TG),total cholesterol(TC),High density lipoprotein(HDL), Low density lipoprotein(LDL),Superoxide Disnutase(SOD)vitality,Malondialdehyde (MDA) content were measured by enzyme dynamics method, The aortic root plaques were observed by oil red O staining.Light microscopy wered adopted to assess the degree of the degre of As lesions of aortic wall and image analysis was performed with computer.ResultsAs compared with the blank control group,shows,1.high dose group could reduce the levels of serum TG,TC,LDL,MDA significantly(p<0.01),but elevate HDL and SOD level significantly(p<0.05). 2. It could also reduce the extent of atherosclerotic lesion and the aorta plaque areas were significantly lower.3.It also could restrain the positive express of NF-κB(p<0.01).ConclusionsThe resulting data provide the ocurrence and the development of atherosclerotic lesions could be inhibited by the Total Flavone of Scutellaria Barbata, by regulating their lipid metabolism, Increasing superoxide dismutase (SOD) activity and reduced malondialdehyde (MDA) formation enhance their antioxidant capacity and reduce the plaque area , NF-κB positive expression.so it could regulating blood lipid ,anti-atherosclerosis and stabilize antheroclerotic plaque,it had great value in treating and Preventing hyperlipedemia, antherosclerosis,ACS,et al.
