Purpose:To study the influence of TangKe decoction of compound Chinese herbs on the function of Activity of endothelial nitric oxide synthase and the release of nitric oxide levels of insulin resistance in type 2 diabetes mellitus rats .Material and method:Rat model of type 2 diabetes mellitus was duplicated by using small dose strepozotocin(STZ) and high caloric diet,and using TangKe decoction of compound chinese herbs and rosiglitazone acted on model rats individually. By using methods of chemical chromometry to determine activity of endothelial nitric oxide synthase . Nitric oxide examination adopts nitrate reductase method.Results:1. Assessment of insulin resistance in model rats:Before the treatment ,model group was significantly lower than normal control group level in insulin sensitive index ( P < 0.01); After the treatment,though the levels of insulin sensitive index of TangKe decoction of compound Chinese herbs group and rosiglitazone group are obviously lower than normal group, they are obviously higher than that of model group(P<0.05).2. Assessment of endothelial nitric oxide synthase activity:After the treatment, TangKe decoction of compound chinese herbs treatment group and the rosiglitazone treatment group were lower than the control group on endothelial nitric oxide synthase activity, but they are obviously higher than that of model group(P<0.05). Insulin sensitive index was significantly positive with inhibitor of endothelial nitric oxide synthase (r=0.63,P<0.01).3. Assessment of nitric oxide levels: Before the treatment, model group was significantly lower than normal control group level in nitric oxide activity ( P < 0.01);after the treatment, TangKe decoction of compound chinese herbs treatment group and the rosiglitazone treatment group were lower than the control group on nitric oxide activity (P<0.05), they are obviously higher than that of model group(P<0.05);and after the treatment, the level of nitric oxide was significantly positive with endothelial nitric oxide synthase activity(r=0.366,P<0.01),and positive with insulin sensitive index(r=0.45,P<0.01).Conclusion:1. TangKe decoction of compound Chinese herbs group can significantly improve the insulin resistance.2. TangKe decoction of compound Chinese herbs group can improve the activity of endothelial nitric oxide synthase in model rats.3. TangKe decoction of compound Chinese herbs group can increase the level of nitric oxide increased by vascular endothelial in model rats.
