Objective:To investigate the influence of exhaustion exercise and contusion to muscle satellite cells and AMPK, TSC2, mTOR in rat.Methods:24 male SD rats were randomly divided into four groups. There were rest control group (C), Exhaustive exercise immediately group (E0),24 h after exhaustive group (E24),48 h after ex haustive group (E48). The rest group and exhaustive exercise rats were anesthetized, taken blo od and the right side of the quadriceps, separated the quadriceps into two parts, one cultured f or experiments immediately, the other saved in a 80 ℃ fridge to detected the content of AMPK, TSC2, mTOR. The contusion group:6 six male S D rats were randomly divided into 3 groups, contusion immediatetely group (DO),24h after c ontusion group (D24),48h after contusion group (D48), anesthetized then taken the right side quadriceps, cultured immediately. Taken another three newborn rats to do the same cell cultur e experiments compare to the contusion group.Results:1, results of muscle satellite cell culture experiment:the rest control group had no evidently muscle satellite cells appeared. Compare to contusion group, the number of muscle s atellite cell of exhaustive exercise group was less, the growth rate was relatively slow. There w as an obvious difference between these two groups. The results show that cells would be prolifer ated in the 5th day, at the 7th day would reach the peak, then, proliferation rate would be de clined. The newborn rat muscle satellite cells had the fastest growth rates, and spindle cells can be seen at the next day of culture afterwards they will considerably proliferate.2, The results of ELISA experiment shows that:The contents of AMPK, TSC2 of EO group in skeletal muscle and in serum content was significantly increased compared with the control group (P<0.01). The content of AMPK, TSC2 in E24 and E48 had no significantly change compared to the control group. On the other hand, the content of mTOR in exhaustive exercise immediate group’s skeletal muscle and serum was decreased obviously compared with the control group, and the difference was significant (P<0.01). But there was no difference among E24, E48 and the control group about the content of mTOR in skeletal muscle and serum. And no differences between E24 group and E48 group.Conclusions:1, Results of cell culture experiments showed that although they both can make skeletal muscle satellite cell activation, compared to exhaustive exercise, the contusion activate the skeletal muscle satellite cell better. But the rest control group hadn’t clearly satellite cell cultured It indicated that skeletal muscle satellite cells of adult rats are in a resting state. Compare with exhaustive exercise group, the muscle satellite cell of contusion group proliferated faster and more obviously. It indicated that contusion must be more severe and stimulate cells more stronger. Under an unstimulated condition, newborn rats had the most satellite cells and more proliferation which suggested that in newborn rats, muscle satellite cells were not in a resting state.2, The content of AMPK、TSC2 in exhaustive exercise immediate group was higher than the other groups, but the content of mTOR was lower with a significantly difference. It indicated that AMPK may be activated by exhaustive exercise, and the phosphorylation of TSC2 at the same time may inhibit the synthesis of mTOR.
