Artemia sinica belongs to Phylum Arthropoda , Class Crustacea , Subclass Branchiopoda, Order Anostaca , Family Artemiidae and Genus Artemia.As small crustaceans,it is distributed in inland salt lakes and coastal salt. Notch signaling pathway is also conserved during the evolution of the interaction between cells to regulate the biological development The Notch signaling pathway functions in the development processes between vertebrate and invertebrates,including cell fate decision,nervous system development and the formation of organ and somites.The signaling pathways of Notch have been deeply identified in Drosophila and mammals. There is a contribute for signaling pathways of Notch in the examination Of Artemia.By blasting a mRNA sequence of Notch in the EST library of Franciscana,we get the same conserved domain which called ANK domain. According to a part of identified sequence of Notch we designed RT-PCR primer sequences of Notch,which were used for RT-PCR and Genome Walking.We obtain 881 long gene fragment in the 3'end.By the method of immunohistochemical to determine the expression sites of Notch during the developmental of Artemia sinica.The results confirmed that uniformly expressed in the eggs of 0h ,5h and 10h; the expression of Notch for Artemia sinica in 15h to 40h,20h and 40h,the expression areas mainly focus in nauplius cephalothorax and there is a little in abdomen; the expression areas for Artemia sinica in 3d mainly focus in enteric canal, abdomen and cephalothorax; the expression areas for Artemia sinica during 5d and 7d mainly focus in Somite,appendage and enteric canal.We take real-time PCR method to analysis the expression quantity of Notch in different development stages. The results shows that the expression quantity of Notch in different development stages is different. Between 0 hours to 10 days,there has two peaks.The relative expression is highest near 20 hours.The expression level is changed with the development of organogenesis.We speculate Notch may play an important role not only in the regulation of embryonic neural development homeostasis,but also involved systemic osmotic, cell growth and differentiation, or other related physiological and biochemical processes.
