| Taurine is one of the richest free amino acid in animal body and takes very important role in all systems. The cysteine sulfinate decarboxylase (CSD) is a key enzyme by which the taurine was synthesized. To investgating the establishment of CSD cRNA probe and its expression in mouse brain cortex, we looked up CSD gene sequences from GENE BANK and designed primers accord to it, and cloned CSD gene from adult mouse brain. We synthesized CSD probe on the basis of this gene by in vitro transcriptional system. Applying this probe in pre-processed mouse brain tissue, and then determinate the positive signal in adult mouse brain sections.Both nerve and hormone play important roles in ovary development and ovary functions. However, the regulatory function of the ovarian sympathetic nerve regarding follicle development in the neonatal rat and the related mechanism is less. To investigating the effects of superior ovary nerve transection of newborn rat on its fertility, 2 days rat's superior ovary nerve was sectioned by surgery. When it came of age, the estrus cycle, conceive percentages, litters, weight of ovary, and ovarian sizes were counted.Experimental results were listed below:ⅰ. Establishment of CSD cRNA probe and its expression in mouse brain cortex.①The total RNA was extracted from adult mouse brain and amplifying the gene according to the designed primers by RT-RCR.②Purifying the target DNA and it was inserted in pGEM-T-Easy vector, then a reconstruct vector was acquired. After that, the reconstruct vector was sequenced. It was found that the orientation of insertion for target DNA is totally right and the target DNA was inserted in vector by trans-direction.③The reconstruct vector was digested with restricted enzyme and purified. It is demonstrated that the reconstructed vector was digested in line shape definitely, and with a high purity.④The labeled probe has a high density compare with control and it is satisfied with application.⑤CSD cRNA signals are broadly expressed in adult mouse brain mantle with high density, and locate in molecular layer, external granular layer, external pyramidal layer, inner granular layer, inner pyramidal layer, and polymorphous cell layer of mouse brain cortex.ⅱ. Effects for transecting of superior ovarian nerve on female rats'reproductivity.It was found that prolonged diestrus stage, decreased estrus stage, decreased litters and conceive percentage, loss of weight of ovary and fewer follicles, as well as increased corpus luteum. All in all, transection of superior ovary nerve of neonatal rat bears a vital responsibility for fertility of rats. |
