Anatomical structure is very important for the understanding of function and pathology of organism, in many a research in biomedical science, the anatomical atlas of organism with micrometer resolution is needed for understanding the micro structure of the organism; and the anatomy atlas of large solid biologic specimen in tissue level is also needed for acquiring more integrated information of organism structure. Hence, it is of great importance to provide a micrometer resolution imaging method which can get the anatomical atlas of large solid biologic specimen.The dissertation is about the research of an optical microscopy for the solid large biology specimen with micrometer resolution, the optical design is introduced by following the process of engineer optical design, which consists of three parts: Firstly, introduces the principal and the location at large, then analyzes the principle of the illumination optical path and the imaging path; Secondly, chooses the optical component, describes the concerned factor of light source, objective, beamsplitter, discuss about the aperture diaphragm which control the optical energy, and the field diaphragm which control the field of view, give the reason why diamond knife has the assistance in the illumination; Finally, combines the optical path and the adjusting, introduce the TDI process and its synchronization error, enhance the contrast of image by limit the TDI area with the rectangle field aperture, analysis how the optical length affect the image acquisition, and improve the illumination quality by adjusting the optical length.In addition, two optical parameters: resolution and distortion are measured. Introduce the theory and the methods of resolution measurement, analyze the theory resolution of the optical path, describes the process of different kinds of PSF, give the test results. Describe how the distortion is measured, and give the conclusion that the distortion of is 0.384%.The research and test proved that the introduced optical microscopy method is capable of getting the anatomical atlas of large solid biologic specimen.
